Rybelsus is a type 2 diabetes medicine for adults which is taken along with exercise and diet to improve blood sugar (glucose) levels. It contains semaglutide. Semaglutide increases the body's insulin levels and decreases its blood sugar levels.
Semaglutide also delays gastric emptying and leads to a reduction in appetite. Semaglutide is, therefore, used for weight loss in specific patients as well. It is also effective in reducing the risk of heart attacks, strokes, and similar cardiovascular issues.

Active Ingredient
Semaglutide is the active pharmaceutical ingredient in Rybelsus tablets. Each Rybelsus tablet contains 14mg of Semaglutide.
Uses of Rybelsus tablets (Semaglutide)
Rybelsus tablets (Semaglutide) are used by adults over the age of 18 for the treatment of diabetes type-2. When taken along with exercise and proper diet, Rybelsus tablets help improve blood sugar (glucose) levels. Rybelsus may also be prescribed for weight loss in specific patients.
Rybelsus tablets are also used to reduce the risk of strokes, heart attacks, and other cardiovascular events.
Mechanism of Action
Rybelsus lowers blood sugar levels in adults with type 2 diabetes mellitus (T2DM). After consumption of food, the stomach releases the incretin hormone, Glucagon-like peptide-1, or GLP-1. GLP-1 reduces blood sugar and stimulates insulin secretion.
Rybelsus is a GLP-1 tablet and works at three levels to achieve a similar result. When the body's blood sugar level is high, Rybelsus encourages the pancreas to release more insulin. It also prevents the liver from making too much sugar or releasing too much sugar into the system.
It reduces appetite as well by slowing the rate at which food leaves the stomach. The stomach thus feels full for a longer duration and craves less food.
Rybelsus Dosage – How to take Rybelsus tablets
Rybelsus tablets should be taken in the dose prescribed by the doctor and for the duration recommended by the doctor.
Rybelsus should be taken first thing in the morning on an empty stomach, at least a half hour before eating or drinking anything. It should be taken with a sip of plain water, no more than 4 ounces of water. It should be swallowed as a whole and should not be crushed, split, or chewed.
Adults should start treatment with a dose of 3mg Rybelsus once a day for 30 days. It must be noted, though, that the 3mg dose is only meant to initiate adults into treatment and does not help control blood sugar levels. After 30 days, dosage should be increased to 7mg Rybelsus tablets, taken once a day. If, after 30 days, blood sugar levels are still not under control, the dosage can be further increased to 14mg Rybelsus tablets.
Missed Dose
If a dose of Rybelsus tablets is missed, it should be skipped for the day. The regular, daily dose of Rybelsus can then be taken the next day. Two Rybelsus tablets should not be taken in one day to compensate for the missed dose.
Overdosage
Emergency medical attention must immediately be sought in the event of overdosage. It may lead to low blood sugar, nausea, and vomiting.
Rybelsus has a long half-life of approximately a week, so along with appropriate treatment for the symptoms exhibited, the patient may also need to be kept under observation for a long period of time.
Storage
Rybelsus should be stored at 68° to 77°F (20° to 25°C); however, excursions are permitted to 59° to 86°F (15° to 30°C). It should be stored and dispensed in the original bottle.
Rybelsus tablets should be stored in the original bottle until the bottle is opened. Once open, the bottle should be stored in a dry area, free from moisture.
Contraindications
Rybelsus tablets should not be taken by anyone with a hypersensitivity or serious allergy to semaglutide or any of Rybelsus’ other ingredients.
Anyone with a history or family history of medullary thyroid cancer (MTC) should also avoid Rybelsus as Rybelsus has been known to cause thyroid C-cell tumours in rodents. Patients with Multiple Endocrine Neoplasia syndrome type 2 (MEN 2) are also advised against taking Rybelsus.
Warnings and Precautions
Before initiating treatment with Rybelsus, it is essential to inform the doctor or healthcare provider of any other medical conditions that the patient may be suffering from, especially the following:
- If the patient has or has ever had any medical issues related to the pancreas or kidneys
- If the patient's vision has ever been affected by diabetes
- If the patient has ever been affected by any kind of stomach or intestinal disorders
A doctor should be consulted immediately if a patient gets a lump or swelling in the neck, develops hoarseness, has trouble swallowing, or experiences shortness of breath. These could be indicative of thyroid cancer.
The healthcare provider should also be made aware of any medicines that the patient may already be taking. This includes prescription medication, over-the-counter medicines, any herbal supplements, or vitamins. These may impact Rybelsus' efficacy or vice versa.
The healthcare provider should especially be informed if the patient is taking any other diabetes medication including insulin or sulfonylureas since it may increase the risk of hypoglycemia (low blood sugar), including severe hypoglycemia.
Rybelsus should not be used for the treatment of type-1 diabetes.
Side effects of Rybelsus tablets
On first taking Rybelsus, side effects may include diarrhoea, nausea, and vomiting.
Other common side effects of Rybelsus may be decreased appetite, stomach pain, and constipation.
Rybelsus can also lead to changes in vision, inflammation of the pancreas, gallbladder and kidney problems, as well as low blood sugar.
Consult a doctor immediately if you suffer from any of these side effects, especially shortness of breath, trouble swallowing, hoarseness, and swelling or lumps in the neck as these could be symptoms of thyroid cancer.
Rybelsus caused thyroid tumours and cancer in animals during studies, but it is undetermined whether it can lead to similar results in humans.
Rybelsus during Pregnancy
Rybelsus should be stopped at least two months before a pregnancy is planned, and a doctor or healthcare provider should be consulted on the best way to keep blood sugar levels in check during this time.
It is not known if Rybelsus can lead to birth defects, miscarriage, or harm the unborn child in any other way, but losing weight has the potential to adversely impact the unborn child, even in overweight patients.
During animal studies, exposure to Rybelsus did sometimes adversely affect the foetus.
Nursing Considerations
Breastfeeding is not recommended in those being treated by Rybelsus.
Do not use this medication without telling your doctor if you are breast-feeding a baby.
