Mesalamine suppository (Generic Canasa) is an aminosalicylate used for the treatment of mild to moderately active ulcerative proctitis. Safety and effectiveness of Generic Canasa suppositories for a duration of more than 6 weeks have not been proved.

Active Ingredient present in Mesacol Suppository
The active ingredient present in Mesacol Suppository (Generic Canasa) is Mesalamine, also known as Mesalazine or 5-Aminosalicylic Acid (5-ASA). Each Mesacol suppository contains 500 mg of Mesalamine.
Uses of Generic Canasa (Mesacol Suppository by Sun Pharma)
This medicine is used to treat ulcerative colitis, proctitis, and proctosigmoiditis. It affects a substance in the body that causes inflammation, tissue damage, and diarrhea. It is also used to prevent the symptoms of ulcerative colitis from recurring.
Mesalamine Suppository Dosage
The dosage of Mesalamine 500 mg suppositories (Generic Canasa) is one rectal suppository once daily at bedtime.
The suppository should remain inserted for one to three hours or longer, if possible. The normal course of therapy is from three to six weeks depending on symptoms and sigmoidoscopic findings.
If a patient misses a dose of Mesalamine Suppository, it should be administered as soon as possible, unless it is almost time for next dose. Patients should not administer two Generic Canasa suppositories at the same time to make up for a missed dose.
How to Use Generic Canasa Suppositories
Use Mesacol suppositories (rectal insert) medicine as directed by your doctor.
How to insert Generic Canasa Suppositories:
Wash your hands before and after using the suppositories. If the suppository is too soft to use, put it in the refrigerator for about 15 minutes. You may also run cold water over it. Remove the wrapper. Moisten the suppository with cold water. Lie down on your side. Insert the pointed end of the suppository into the rectum. Use your finger to push it in completely.
Do not handle the suppository too much. This will cause the suppository to melt before it can be inserted. For best results, keep the suppository inserted for at least 1 to 3 hours.
Use Mesacol Suppository for the full course of treatment. Keep using it even if you feel better in a few days.
Take a missed dosage as soon as possible. If it is almost time for your next dosage, skip the missed dose and go back to your regular dosing schedule. Do not use 2 doses at once.

Generic Canasa (Mesalamine Suppository) Contraindications
Generic Canasa (Mesalamine Suppository) is contraindicated in patients with a hypersensitivity to Mesalamine (5-aminosalicylic acid) or to the inactive ingredients present in the suppository vehicle (saturated vegetable fatty acid esters), or to salicylates (including aspirin).
Mesalamine Suppositories Side effects
The most common Mesalamine suppositories side effects occurring in more than 1% of treated patients are: dizziness, rectal pain, fever, rash, acne and colitis.
Get emergency medical help if you have any of these signs of an allergic reaction to Mesacol Suppository: hives; difficulty breathing; swelling of your face, lips, tongue, or throat.
Stop taking this medicine and call your doctor at once if you have a serious side effect such as severe stomach pain, cramping, fever, headache, and bloody diarrhea.
Less serious side effects of Mesalamine Suppositories may include:
• mild nausea, vomiting, stomach cramps, diarrhea, gas
• fever, sore throat, or other flu symptoms
• constipation
• headache or dizziness
• tired feeling
• skin rash

Overdose symptoms may include ringing in your ears, dizziness, headache, confusion, drowsiness, sweating, shortness of breath, vomiting, and diarrhea.
Warnings & Precautions When Using Mesalamine Suppositories
Mesalamine Suppositories may cause dizziness. The effect may be worse if you take it with alcohol or certain medicines. Use this medicine with caution. Do not drive or perform other possibly unsafe tasks until you know how you react to it.
Mesacol suppository may stain clothing, other fabrics, or surfaces. Avoid contact with your clothes, other fabrics, or surfaces.
Salicylates have been linked to a serious illness called Reye syndrome. Do not give this medicine to a child, or teenager, or a person who has the flu, chickenpox, or a viral infection. Contact your doctor with any questions or concerns.
Lab tests, including kidney function, liver function, or complete blood cell counts, may be performed while you use this medicine. These tests may be used to monitor your condition or check for side effects. Be sure to keep all doctor and lab appointments.
Use this medicine with caution in the elderly; they may be more sensitive to its effects, especially blood problems. It should be used with extreme caution in children; safety and effectiveness in children have not been confirmed.
Do not use Mesacol Suppository if you are allergic to any ingredient in this medicine, nonsteroidal anti-inflammatory drugs (NSAIDs) (eg, ibuprofen, naproxen), or salicylates (eg, aspirin).
Tell your doctor if you have any medical conditions, especially if any of the following applies to you:
• if you are pregnant, planning to become pregnant, or are breast-feeding
• if you are taking any prescription or nonprescription medicine, herbal preparation, or dietary supplement
• if you have allergies to medicines (including sulfasalazine), foods, or other substances
• if you have had a stroke, bleeding in the brain, or an aneurysm
• if you have a history of liver or kidney problems, pancreas problems (eg, pancreatitis), Kawasaki syndrome, or a rheumatic (inflammatory) disease (eg, rheumatoid arthritis)
• if you have a history of heart problems (eg, inflammation of the sac around your heart)
• if you have the flu or chickenpox, or if you have recently received a live vaccine (eg, varicella, influenza)
Other medicines may have adverse reactions when combined with the active ingredient in this medicine. Tell your health care provider if you are taking any other medicines, especially any of the following:
• Ginkgo biloba extract
• Corticosteroids (eg, Prednisone®)
• Carbonic anhydrase inhibitors (eg, acetazolamide), insulin, meglitinide antidiabetics (eg, Repaglinide®), sulfonylureas (eg, Glipizide®), or valproic acid
• Angiotensin-converting enzyme (ACE) inhibitors (eg, Enalapril®), anticoagulants (eg, Warfarin®), beta-blockers (eg, Propranolol®), probenecid, spironolactone, or sulfinpyrazone
This may not be a complete list of all interactions that may occur. Ask your health care provider if this medicine may interact with other medicines that you take. Check with your health care provider before you start, stop, or change the dosage.
Mesalamine Suppositories may cause you to become sunburned more easily. Avoid the sun, sunlamps, or tanning booths until you know how you react to the medicine. Use a sunscreen or wear protective clothing if you intend to be outside for a longer period of time.
Mesalamine Suppository (Generic Canasa) During Pregnancy
Mesalamine Suppository (Generic Canasa) have been classified by the US FDA as Pregnancy Category B. Reproduction studies performed in rats at doses up to 320 mg/kg/day (about 1.7 times the recommended human intra-rectal dose, based on body surface area) have shown no proof of impaired fertility or harm to the fetus due to Mesalamine.
There are, however, no sufficient and well-controlled studies conducted in pregnant women. Since animal reproduction studies are not always predictive of human response, Mesalamine suppository should be used during pregnancy only if clearly needed.
Generic Canasa may be harmful to an unborn baby. Tell your doctor if you are pregnant or plan to become pregnant during treatment.
Mesalamine Suppository (Mesacol) During Breastfeeding
Low concentrations of Mesalamine and higher concentrations of its N-acetyl metabolite have been proved to be present in human breast milk. It is not known at present whether this medicine could harm a nursing baby. Do not use Mesalamine suppository without informing your doctor if you are breastfeeding a baby.
